ABSTRACT

O objetivo deste trabalho foi avaliar macro e microscopicamente a atividade cicatrizante da Sphagneticola trilobata em feridas cutâneas induzidas em ratos, a partir da aplicação de creme contendo extrato hidroalcoólico bruto de folhas da planta. A análise fitoquímica apresentou terpenos e flavonoides como compostos majoritários. Sessenta ratos foram divididos em três grupos experimentais (n=20): grupo tratado (GT), grupo controle (GC) e grupo controle absoluto (GCA). Quatro feridas excisionais de 0,8cm de diâmetro foram realizadas no dorso dos animais, tratadas diariamente e avaliadas nos tempos três, sete, 14 e 21 dias de pós-operatório (PO) quanto à contração e à avaliação macroscópica, morfo-histológica e morfo-histométrica. Macroscopicamente, não houve diferença estatística na contração das feridas entre os grupos testados. Na avaliação morfológica e na morfométrica, o GT apresentou menor concentração de células inflamatórias, maior e melhor preenchimento do tecido de granulação pelas fibras colágenas e melhor vascularização das feridas. Não houve diferença entre o GC e o GCA. Conclui-se que o creme à base do extrato hidroalcoólico bruto das folhas de Sphagneticola trilobata contribui positivamente para o processo de cicatrização das feridas em pele de ratos.(AU)


The objective of this work was to macro and microscopically evaluate the healing activity of Sphagneticola trilobata in rat-induced skin wounds by applying cream containing crude hydroalcoholic extract from plant leaves. The phytochemical analysis showed terpenes and flavonoids as major compounds. Sixty rats were divided into three experimental groups (n=20): treated group (GT), control group (CG) and absolute control group (GCA). Four 0.8cm diameter excision wounds were performed on the back of the animals, treated daily and evaluated at the three, seven, 14 and 21 postoperative days (PO) for contraction, macroscopic, morphologic and morphologic evaluation. The TG presented smaller scar area at 21 postoperative days (P<0.05). In the morphological and morphometric evaluation, the WG presented lower inflammation, greater and better filling of granulation tissue by collagen fibers and better wound vascularization. There was no difference between GC and GCA. It was concluded that the cream based on the crude hydroalcoholic extract of Sphagneticola trilobata leaves contribute positively to the healing process of the skin wounds of rats.(AU)

ABSTRACT

Several isatin derivatives have shown important biological activities, which have attracted interest from researchers. For this reason, the present study aimed to evaluate the anti-inflammatory and antinociceptive effects of the isatin derivative (Z)-2-(5-chloro-2-oxoindolin-3-ylidene)-N-phenyl-hydrazinecarbothioamide (COPHCT) in mice. Three doses of this compound were tested: 1.0, 2.5, and 5.0 mg/kg. The anti-inflammatory activity was assessed using the carrageenan-induced paw edema model and the zymosan-induced air pouch model. The evaluation of the antinociceptive effect was performed through the formalin test and the acetic acid-induced abdominal writhing test. The paw edema assay demonstrated that all doses of the compound showed a significant reduction of the edema in the second hour evaluated, but a better response was observed in the fourth hour. The zymosan-induced air pouch model indicated that the compound, in all doses, significantly reduced leukocyte migration and total protein concentration levels. In the formalin test, the doses 1.0, 2.5, and 5.0 mg/kg of COPHCT showed activity only in the second phase, with reduction in paw pain time of 73.61, 79.46, and 73.85%, respectively. The number of abdominal writhings decreased with the increasing dose, but only 5.0 mg/kg COPHCT exhibited a significant response, with a reduction of 24.88%. These results demonstrated the ability of this compound to interfere in the anti-inflammatory activity of edema, vascular permeability, and cell migration. In addition, its possible antinociceptive effect may be related to the dose used.

ABSTRACT

Avaliou-se o efeito da adição do ácido linoleico conjugado (CLA) ao meio de cultivo in vitro na viabilidade pós-vitrificação de embriões F1 Holandês x Zebu. Foram utilizados três meios de cultivo: controle (n=340 oócitos): meio SOF e soro fetal bovino (SFB), sem o CLA; SFB+CLA (n=359 oócitos): meio SOF, SFB e CLA; CLA (n=339 oócitos): meio SOF e CLA, sem o SFB. Todos os blastocistos produzidos foram submetidos à vitrificação, pelo método de Open Pulled Straw. Quinze blastocistos de cada tratamento foram fixados para quantificação lipídica por coloração com Sudan Black B. Para avaliar a viabilidade embrionária, foi observada a capacidade de reexpansão e eclosão pós-aquecimento dos embriões (controle=27; SFB+CLA=30; CLA=17). Foram realizadas transferências em um ou dois embriões por receptora para avaliação da sobrevivência in vivo: T1 [receptoras que receberam um blastocisto (n=17 embriões, sendo controle=5, SFB+CLA=6 e CLA=6)]; T2 [receptoras que receberam dois blastocistos, (n= 54 embriões, sendo controle=18, SFB+CLA=14 e CLA=22)]. Não houve diferença nas taxas de clivagem (62,1%; 74,0%; 74,0% para controle; SFB+CLA; CLA, respectivamente), produção de blastocistos em relação aos clivados (59,7%; 47,7%; 38,3% para controle; SFB+CLA; CLA, respectivamente) e produção de blastocistos em relação ao total de oócitos (37,1%; 35,4%; 28,3% para controle; SFB+CLA; CLA, respectivamente) (P>0,05). Houve diminuição de gotículas lipídicas nos embriões cultivados em meio suplementado com CLA em relação aos embriões cultivados na presença do SFB e na ausência do CLA (P<0,05). A taxa de reexpansão foi maior no grupo controle (70,4%) em relação ao CLA (47,1%) e menor no grupo SFB+CLA (43,3%) (P<0,05). O CLA foi eficaz em reduzir a deposição de lipídeos intracitoplasmáticos nas células embrionárias, porém não houve diferença de viabilidade após a desvitrificação dos embriões.(AU)


The effect of adding conjugated linoleic acid (CLA) to the culture media on the viability after cryopreservation of F1 Holstein X Zebu embryos was evaluated. Three different culture media were tested: control (n = 340 oocytes): SOF medium and fetal bovine serum (FBS) without the CLA; FBS + CLA (n = 359 oocytes): SOF, FBS and CLA; CLA (n = 339 oocytes): SOF and CLA without the FBS. The produced blastocysts were subjected to vitrification, by the Open Pulled Straw method. Fifteen blastocysts per treatment were fixed for lipid quantification by staining with Sudan Black B. Embryo re-expansion and hatching capability were used to assess viability (control = 27; FBS + CLA = 30; CLA = 17). Transfers of one or two embryos to recipients were performed to evaluate in vivo survival: T1 [recipients that received one blastocyst (n=17 embryos, Control=5, FBS+CLA=6 and CLA=6)]; T2 [recipients that received two blastocysts (n =54 embryos, Control=18, FBS+CLA=14 and CLA=22)]. There was no difference in cleavage rate (62.1%; 74%; 74% for Control; FBS + CLA, CLA, respectively), blastocyst production in relation to the cleaved structures (59.7%; 47.7%; 38 3% for Control; FBS + CLA, CLA, respectively) and blastocyst production relative to the total oocytes (37.1%, 35.4%, 28.3% for Control; FBS + CLA, CLA, respectively) between treatments (P> 0.05). A reduction of lipid droplets was observed in embryos cultured in medium supplemented with CLA compared to embryos cultured in the FCS in the absence and presence of CLA (P <0.05). The reexpansion rate was higher in the Control group (70.4%) compared to the CLA (47.1%) and lowest for FBS+CLA (43.3%) (P<0.05). The hatching rates were similar among treatments, 42.1%; 23.1%; 25% for control; SFB + CLA; CLA respectively (P>0.05). Only one pregnancy was observed in early and confirmatory diagnosis, as the result of a Control group embryo transfer. Although embryos cultured with CLA have shown smaller intracytoplasmic lipid content, no difference was observed in viability following vitrification between treatments.(AU)

De um total de 2.088 ruminantes necropsiados no Laboratório de Patologia Animal da Universidade Federal de Campina Grande durante o período de janeiro de 2003 a dezembro de 2015, 803 (38,45%) eram bovinos, 651 (31,18%) ovinos e 634 (30,37%) caprinos. Desses, dois caprinos (0,31%), um ovino (0,15%) e um bovino (0,12%) foram diagnosticados com ruptura de artéria aorta devido à presença de aneurismas, correspondendo a 0,19% das causas de morte em ruminantes. O caso 1 ocorreu em um caprino que apresentava bom estado corporal e foi encontrado morto pelo proprietário; o caso 2, em um caprino com diagnóstico clínico presuntivo de linfadenite caseosa; o caso 3, em um ovino que foi encontrado morto pelo proprietário; e o caso 4, em um bovino com histórico de timpanismo recidivante. Em todos os casos, a morte ocorreu rapidamente por choque hipovolêmico. Os principais achados macroscópicos na necropsia foram hemotórax ou hemoperitônio e a ruptura na artéria aorta. Nos caprinos, abscessos foram visualizados microscopicamente e estavam associados à presença de bactérias, o que sugere infecção por Corynebacterium pseudotuberculosis. No ovino, não foram observadas lesões que pudessem elucidar a causa do aneurisma, por isso foi considerado de provável origem idiopática. No bovino, não foi possível estabelecer a causa, mas a presença do aneurisma dificultava a eructação dos gases da fermentação ruminal, caracterizando um quadro de timpanismo secundário. Aneurismas ocorrem de forma esporádica em caprinos, ovinos e bovinos e devem ser considerados como uma importante causa de morte súbita.(AU)


From a total of 2,088 necropsied ruminants in the Laboratory of Animal Pathology of Federal University of Campina Grande during the period from January 2003 to December 2015, 803 (38.45%) were cattle, 651 (31.18%) sheep, and 634 (30.37%) goats. Of these, two goats (0.31%), one sheep (0.15%) and one bovine (0.12%) were diagnosed with rupture of aorta due to the presence of aneurysms, corresponding to 0.19% of causes of death in ruminants. Case 1 occurred in a goat that had good body condition and was found dead by the owner, case 2 occurred in a goat that had clinical presumptive diagnosis of caseous lymphadenitis, case 3 occurred in a sheep who was found dead by the owner, and case 4 occurred in a cow with a history of recurrent bloating. In all cases, death occurred rapidly by hypovolemic shock. The macroscopic findings at necropsy were hemothorax or hemoperitoneum and the rupture in the aorta artery. In goats, abscesses were viewed microscopically and they were associated with the presence of bacteria, suggesting for Corynebacterium pseudotuberculosis infection. In the sheep, lesions that could explain the cause of the aneurysm were not observed, so we consider the aneurisma as idiopathic. In the cow, it was not possible to establish the cause, but the presence of the aneurysm hampered eructation of ruminal gases, causing secondary bloat. Aneurysms occur sporadically in goats, sheep and cattle and must be regarded as an important cause of sudden death.(AU)

Abstract Phenology and reproductive biology of cultivated species are important for the comprehension of the requirements for fruit and seed production and the management of pollinators. This study aimed to characterise the phenology, reproductive biology and growing degree days of the grapevine ‘Isabel’ (Vitis labrusca) in northeastern Brazil during January 2011 (P1), Augst 2011 (P2), April 2012 (P3) and August 2012 (P4). We recorded the duration (days) of the phenological stages, pruning (P), woolly bud (W), budburst (B), inflorescence development (ID), flowering (F), ripening (R) and harvest (H). We analysed the floral biology, the sexual system and the breeding system. We measured the growing degree days (GDD) required to reach the subperiods P-B, B-F and F-H. The periods P1, P2, P3 and P4 lasted for 116, 125, 117 and 130 days, respectively. The number of days of harvest were similar in the same dry (P1 and P3) and rainy (P2 and P4) periods. All the periods that we recorded were shorter than those observed in other regions of Brazil, which may be attributable to the mean temperature and carbohydrate metabolism. The flowers are green, hermaphroditic, with an odour of mignonette, low pollen viability and autogamous. The base temperature of 10°C was considered the most adequate for the subperiods as has been documented for other grape varieties in Brazil. Thus, temperature was also the most adequate for the cycles, presenting a smaller standard deviation (0.119, 0.147, 0.156 and 0.153 to P1, P2, P3 and P4, respectively) when compared to a base temperature of 12°C (0.122, 0.158, 0.165 and 0.160 to P1, P2, P3 and P4, respectively). The higher and the lower observed GDD were 1972.17 and 1870.05, respectively, both above the values recorded in other parts of Brazil for same variety. The phonological results, including knowledge of growing degree days, are important to the planning of cultures at the study site and in other regions that have similar climatic conditions and make it possible to pre-determine the harvest.


Resumo Fenologia e biologia reprodutiva de espécies cultivadas são importantes para a compreensão dos requerimentos para a produção de frutos e sementes, bem como para o manejo de polinizadores. O presente estudo objetivou caracterizar a fenologia, a biologia reprodutiva e a exigência térmica (graus-dias) da videira “Isabel” (Vitis labrusca) no nordeste do Brasil, durante janeiro/2011 (P1), Agosto/2011 (P2), Abril/2012 (P3) e Agosto/2012 (P4). Analisamos a duração (dias) dos estádios fenológicos poda (PO), gema-algodão (GA), brotamento (BR), aparecimento da inflorescência (AI), florescimento (FL), início da maturação (IM) e colheita (CO). Analisamos a biologia floral, o sistema sexual e o sistema reprodutivo. As exigências térmicas foram obtidas em termos de graus-dia (GD) necessários para atingir os subperíodos PO-BR, BR-FL e FL-CO. A duração do ciclo foi de 116, 125, 117 e 130 dias para as épocas P1, P2, P3 e P4 respectivamente, sendo o número de dias de colheita semelhantes no período seco (P1 e P3) e chuvoso (P2 e P4). Todos os períodos analisados foram mais curtos do que aqueles observados em outras regiões do Brasil, o que pode ser atribuído à temperatura média e ao metabolismo de carboidratos. As flores são verdes, hermafroditas, com odor almiscarado, baixa viabilidade polínica e autógama. A temperatura base de 10ºC foi considerada a mais adequada para os subperíodos e tem sido documentada para outras variedades de uva no Brasil; portanto, a temperatura foi também a mais adequada para os ciclos, apresentando um desvio padrão menor (0,119; 0,147; 0,156 e 0,153 para P1, P2, P3 e P4, respectivamente), comparado com uma temperatura base de 12ºC (0,122; 0,158; 0,165 e 0,160 para P1, P2, P3 e P4, respectivamente). O maior é o menor GDD observado foram 1972,17 e 1870,05, respectivamente. Os resultados fenológicos, incluindo o conhecimento dos graus dias, são importantes para o planejamento de culturas no local e em outras regiões com condições climáticas semelhantes, tornando possível pré-determinar a colheita.

ABSTRACT

Objetivou-se avaliar os efeitos de antioxidantes suplementados às dietas de galos de matrizes pesadas em idade avançada sobre as características reprodutivas. Foram utilizados galos da linhagem Cobb com 47 semanas de idade. Estes foram distribuídos em quatros tratamentos, de acordo com a adição de antioxidante à dieta, sendo: A = grupo controle (sem suplementação antioxidante); B = suplementação de 9ppm de citranaxantina; C = suplementação de 6ppm de cantaxantina e D = 150ppm de vitamina E. Foram avaliadas as características seminais, tais como volume, vigor, motilidade e concentração, a composição de ácidos graxos do sêmen (saturados, insaturados, monoinsaturados, poli-insaturados, ômega 3, ômega 6 e ômega 9) e a correlação entre o peso dos testículos e o peso dos galos. Não foram observados efeitos das substâncias antioxidantes testadas sobre os parâmetros reprodutivos avaliados. Os antioxidantes avaliados não influenciam as respostas reprodutivas de machos reprodutores de matrizes pesadas em idade avançada.(AU)


This study aimed to evaluate the effect of antioxidant supplementation in diets of aging broiler breeder roosters on reproductive characteristics. Cobb roosters were divided into four groups, according to antioxidant addition, where: A = control group (without antioxidant supplementation); B = 9ppm of citranaxanthin; C = 6ppm of canthaxanthin, and D = 150ppm of vitamin E. Seminal characteristics (volume, force, motility and concentration), semen fatty acid composition (saturated, unsaturated, monounsaturated, polyunsaturated, ω3, ω6 and ω9), and the correlation between testis weight with rooster weight were evaluated. No effects of the antioxidants tested were observed. The antioxidants do not influence the reproductive responses of aging male broiler breeder.(AU)

ABSTRACT

The most important role played by the enzyme Glucose-6-Phosphate Dehydrogenase (G6PD) in erythrocyte metabolism is in generating energy and reducing power used to protect the cell against oxidative attack. G6PD deficiency is the erythroenzymopathy that most frequently causes hemolytic anemia, and more than 130 molecular variants have already been identified. The aim of this study was to analyze the genetic mutations in the G6PD-deficient adult males in the population of the region of Araraquara, São Paulo State. Out of 5087 male blood donors, 89 were deficient for G6PD, as confirmed by assaying the enzyme activity and electrophoresis on cellulose acetate. Thus, a frequency of 1.75% of G6PD-deficient patients was found, this value being similar to other investigations in São Paulo state. Molecular analysis was performed by amplification of genomic DNA with specific primers and digestion with restriction enzymes. In 96.6% of the patients, the G6PD A¯ variant was observed, with mutations at residues 376(A-G) and 202(G-A). Mean G6PD specific activity among the patients was 1.31 IU.g Hb-1.min-1 at 37ºC, that is 10.8% of the normal activity of the G6PD B enzyme. The variant forms G6PD A¯680(G-T) and 968(T-C) were not found. In 3.4% of the deficient individuals, the G6PD Mediterranean variant was found, with a mutation at 563(C-T). In these cases,mean enzymatic activity was 0.25 IU.g Hb-1.min-1 at 37ºC, or 2.1% of the enzymatic activity of G6PD B. A importância da enzima Glicose-6-fosfato desidrogenase (G6PD) no metabolismo eritrocitário está na obtenção de energia calórica e redutora para a proteção celular contra agressões oxidativas. A deficiência de G6PD é a eritroenzimopatia que causa mais frequentemente anemia hemolítica, com mais de 130 variantes moleculares identificadas. O objetivo deste estudo foi realizar a análise molecular da deficiência de G6PD em uma população masculina adulta da região de Araraquara, SP, para a identificação das mutações genéticas. Nos 5087 doadores de sangue do sexo masculino pesquisados, foram encontrados 89 deficientes de G6PD, confirmados pela determinação da atividade enzimática e eletroforese em acetato de celulose, com frequência de 1,75%, valores semelhantes aos encontrados por outros pesquisadores no Estado de São Paulo. A análise molecular realizada pela amplificação do DNA genômico com iniciadores específicos e digestão com enzimas de restrição, demonstrou que 96,6% dos deficientes apresentaram a variante G6PD A¯, com as mutações 376(A-G) e 202(G-A) e atividade enzimática média de 1,31 UI.g de Hb-1.min-1 a 37ºC, correspondendo a 10,8% da atividade enzimática da enzima normal G6PD B. Não foram encontradas as formas variantes G6PD A¯ 680(G-T) e 968(T-C). Em 3,4% dos indivíduos deficientes, foi encontrada a variante G6PD Mediterrânea, mutação 563(C-T) e atividade enzimática média de 0,25 UI.g de Hb-1.min-1 a 37ºC, correspondendo a 2,1% da atividade enzimática da G6PD B. ABSTRACT

The effects of subarachnoid administration of iohexol on intracranial hemodynamic in dogs anesthetized with propofol or sevoflurane were evaluated. Thirty adult animals (10.9±2.9kg) were distributed into two groups: PG, where propofol was used for induction (10±0.5mg/kg), followed by a continuous rate infusion at 0.55±0.15mg/kg/hour, and SG, where sevoflurane was administered for induction (2.5 MAC) and for anesthetic maintenance (1.5 MAC). A fiberoptic catheter was implanted on the right superficial cerebral cortex to monitor intracranial pressure (ICP). After 30 minutes, cerebrospinal fluid (CSF) was collected at the cisterna magna and iohexol was injected. The measurements were performed before CSF collection (TA), after the iohexol injection (T0), and at 10-minute intervals (T10 to T60). Intracranial pressure decreased at T0 in SG. Cerebral perfusion pressure at T0 was higher than at TA, T50 and T60 in PG, but in SG, the mean value at T0 was higher than the ones from T20 to T60. Mean arterial pressure at T0 was higher than at TA in PG, while in SG, the values from T20 to T60 were lower than at T0. The heart rate at T60 was lower than at T0 in PG. Cardiac output at TA was lower than at T60 in SG. The cerebrospinal fluid collection and administration of iohexol promoted decrease in intracranial pressure in sevolflurane-anesthetized dogs and increase in cerebral perfusion pressure in propofol-anesthetized dogs.


Avaliaram-se os efeitos da administração subaracnóidea de iohexol sobre a hemodinâmica intracraniana em cães anestesiados com propofol ou sevofluorano. Trinta e dois animais (10,9±2,9kg) foram distribuídos em dois grupos: no GP, o propofol foi usado para indução, 10±0.5mg/kg, seguido por infusão contínua, 0,55±0,15mg/kg/min; no GS, o sevofluorano foi administrado para indução, 2,5 CAM, e manutenção, 1,5 CAM, da anestesia. O cateter de fibra óptica foi implantado na superfície direita do córtex cerebral para monitorar a pressão intracraniana (PIC). Após 30 minutos, o fluido cerebroespinhal (FCS) foi coletado da cisterna magna e o iohexol injetado. As mensurações ocorreram antes da coleta do FCS (TA), depois da injeção de iohexol (T0) e em intervalos de 10 minutos (T10 a T60). A pressão intracraniana diminuiu em T0 no GS. A pressão de perfusão cerebral em T0 foi maior que em TA, T50 e T60 no GP, mas no GS, a média em T0 foi maior que as de T20 a T60. A pressão arterial média em T0 foi maior que em TA no GP, enquanto no GS, de T20 a T60, as médias foram menores que em T0. A frequência cardíaca em T60 foi menor que em T0 no GP. O débito cardíaco em TA foi menor que em T60 no GS. A coleta do fluido cerebroespinhal e a administração do iohexol promoveram a diminuição da pressão intracranina em cães anestesiados com sevofluorano e aumento da pressão de perfusão cerebral em cães anestesiados com propofol.

ABSTRACT

The objective of this study was to assess the prevalence of barriers to interferon treatment in a population of HIV/HCV coinfected patients. A cross-sectional study was conducted at two AIDS Outpatient Clinics in Brazil. The study included all HIV infected patients followed at these institutions from January 2005 to November 2007. Medical records of 2,024 HIV-infected patients were evaluated. The prevalence of anti-HCV positive patients among them was 16.7 percent. Medical records of HCV/HIV coinfected patients were analyzed. 189 patients with the following characteristics were included in our study: mean age 43 years; male gender 65 percent; former IDUs (52 percent); HCV genotype 1 (66.4 percent); HCV genotype 3 (30.5 percent); median CD4+ T cell count was 340 cells/mm³. Among 189 patients included in the analyses, only 75 (39.6 percent) were considered eligible for HCV treatment. The most frequent reasons for non-treatment were: non-compliance during clinical follow-up (31.4 percent), advanced HIV disease (21.9 percent), excessive alcohol consumption or active drug use (18.7 percent), and psychiatric disorders (10.1 percent). CONCLUSIONS: In Brazil, as in elsewhere, more than half of HIV/HCV coinfected patients (60.4 percent) have been considered not candidates to received anti-HCV treatment. The main reasons may be deemed questionable: non-adherence, drug abuse, and psychiatric disease. Our results highlight the importance of multidisciplinary teams to optimize the access of coinfected patients to HCV treatment.

ABSTRACT

O objetivo deste estudo foi analisar os fatores de risco relacionados às doenças cardiovasculares (FRDCV), em idosos diabéticos do tipo 2, através de um estudo observacional com delineamento transversal, tendo sido realizado com 100 idosos atendidos no Centro Regional de Reabilitação de Araraquara (CRRA), São Paulo, Brasil, de março a dezembro de 2004. Houve predominância de indivíduos casados, sexo feminino, brancos, com baixa renda e baixa escolaridade. Quanto aos hábitos e estilo de vida, predominaram os que mantinham dietas adequadas, sedentários, não fumantes e não etilistas. Entre os 100 indivíduos estudados, 42% apresentaram sobrepeso, 42% obesidade, 71% valores inadequados de circunferência abdominal e 84% de relação cintura-quadril de acordo com o sexo. Em relação aos FRDCV, 54,0% apresentaram hipertensão arterial sistêmica (HAS), 54,0% com hipercolesterolemia, 51,0% de hipertrigliceridemia, 84% com valores elevados de LDL-colesterol, 59% com níveis reduzidos de HDL-colesterol, 78,0% com níveis elevados de glicemia de jejum, 76,0% com níveis elevados de hemoglobina glicada e 57,0% de fibrinogênio. Os resultados demonstraram uma frequência elevada de FRDCV com diferenças em relação ao sexo e a idade.


The aim of this study was to analyze the risk factors related to the cardiovascular diseases (CVRF) in elderly type 2 diabetics. A cross-sectional observational study was carried out on 100 elderly patients attending the Rehabilitation Center of Araraquara (CRRA), São Paulo State, Brazil, from March to December, 2004. The majority were married, female, white, with a low income and low educational level. Regarding habits and style of life, the subjects had an adequate diet, were sedentary, non-smoking and non-drinking. In the population of 100, 42% were overweight, 42% obese, 71% had abovenormal waist measurements and 84% high waist-to-hip ratios. Concerning the CVRF, it was observed that more than half had hypertension, hypercholesterolaemia and hypertriglyceridaemia. 84% had high values of LDLcholesterol and 59% HDL-cholesterol levels below the reference values, 78% high levels of fasting glycemia, 76% glycohemoglobin and 57% fibrinogen and thus subject to cardiovascular risk. The results showed a highfrequency of cardiovascular risk factors, differing according to sex and the age.

ABSTRACT

Pemphigus is an inflammatory autoimmune disorder of the skin. Nitric oxide (NO) is an inflammatory mediator linked to a variety of physiological and pathophysiological phenomena that include skin tumors, psoriasis, urticaria, and atopic dermatitis. Inflammatory cells present in pemphigus lesions are important sources of NO production. We investigated whether NO is involved in pemphigus. A prospective cohort study was conducted at the Dermatology Service of the Hospital Universitário Walter Cantídio of the Federal University of Ceará. All patients seen at the outpatient clinic between August 2000 and July 2001, with a clinically and histologically confirmed diagnosis of pemphigus were included. The median age was 42.5 years (range: 12-69 years) with a male to female ratio of 3:2. Total serum nitrite levels, used as a marker for NO production, were determined by the Griess reaction. Skin biopsies from pemphigus and breast surgery (control) patients were used for the detection of the inducible NO synthase (iNOS) by immunohistochemistry. Twenty-two (22) patients with pemphigus and eight (8) controls who did not differ in demographic characteristics were included. Total serum nitrite levels were significantly higher (>7 æmol/L) in pemphigus patients compared to controls (<6 æmol/L), regardless of the severity of the clinical activity of pemphigus (P < 0.0001). All pemphigus biopsies presented increased immunostaining for iNOS that was not detected in normal skin samples. These data are the first to demonstrate that pemphigus patients display increased serum NO levels that are associated with increased iNOS expression in the affected skin.

ABSTRACT

The pathogenesis of nonsteroidal anti-inflammatory drug (NSAID) enteropathy is a complex process involving the uncoupling of mitochondrial oxidative phosphorylation and inhibition of cyclooxygenase (COX). Rofecoxib, a selective inhibitor of COX-2, has shown less gastric damage, but the same beneficial effect is not clear in the case of the small bowel. Fifty-seven male Wistar rats (250-350 g) were divided into three groups (N = 19 each) to evaluate the effect of this NSAID on the rat intestine. The groups received 2.5 mg/kg rofecoxib, 7.5 mg/kg indomethacin or water with 5 percent DMSO (control) given as a single dose by gavage 24 h before the beginning of the experiment. A macroscopic score was used to quantify intestinal lesions and intestinal permeability was measured using [51Cr]-ethylenediaminetetraacetic acid ([51Cr]-EDTA). The extent of intestinal lesion, indicated by a macroscopic score, was significantly lower when rofecoxib was administered compared to indomethacin (rofecoxib = 0.0 vs indomethacin = 63.6 ± 25.9; P < 0.05) and did not differ from control. The intestinal permeability to [51Cr]-EDTA was significantly increased after indomethacin (control = 1.82 ± 0.4 vs indomethacin = 9.12 ± 0.8 percent; P < 0.0001), but not after rofecoxib, whose effect did not differ significantly from control (control = 1.82 ± 0.4 vs rofecoxib = 2.17 ± 0.4 percent; ns), but was significantly different from indomethacin (indomethacin = 9.12 ± 0.8 vs rofecoxib = 2.17 ± 0.4 percent; P < 0.001). In conclusion, the present data show that rofecoxib is safer than indomethacin in rats because it does not induce macroscopic intestinal damage or increased intestinal permeability.

Objetivo.Presentar los pasos fundamentales para incorporar en la práctica clínica los conceptos de la medicina basada en evidencias.Método.Búsqueda en la base de datos Medline de textos relacionados al tema,revisión de varios libros de epidemiología clínica y artículos seleccionados del archivo personal del autor.Resultados.La medicina basada en evidencias reconoce dos fuentes principales de información para la toma de decisión clínica:el paciente individual y la indagación clínica-epidemiológica.A través de esta medicina,la experiencia clínica individual debe ser integrada con las mejores experiencias externas provenientes de la investigación sistemática.El análisis de las evidencias externas requiere un conjunto de nuevos conocimientos y habilidades para el médico actual.Estas habilidades capacitan al médico a adquirir autonomía en el juicio crítico de la calidad de las informaciones disponibles que se utilizarán para disminuir las incertidumbres de sus decisiones clínicas sobre la salud de los pacientes.La práctica de la medicina basada en evidencias compromete responder a algunos desafíos presentes en la práctica a saber:1)cómo mantenerse actualizado frente a la creciente disponibilidad de informaciones en salud.2)cómo seleccionar eficientemente las mejores fuentes de información.3)cómo evaluar críticamente las informaciones disponibles.4)cómo sintetizar las evidencias encontradas.5)cómo integrar las evidencias seleccionadas y la experiencia clínica en el manejo de los problemas de los pacientes

ABSTRACT

Glucose-6-phosphate dehydrogenase (G6PD) activity and the affinity for its substrate glucose-6-phosphate were investigated under conditions similar to the physiological environment in terms of ionic strength (I: 0.188), cation concentration, pH 7.34, and temperature (37oC). A 12.4, 10.4 and 21.4 percent decrease was observed in G6PD B, G6PD A+ and G6PD A- activities, respectively. A Km increase of 95.1, 94.4 and 95.4 percent was observed in G6PD B, G6PD A+ and G6PD A-, respectively, leading to a marked decrease in affinity. In conclusion, the observation of the reduced activity and affinity for its natural substrate reflects the actual pentose pathway rate. It also suggests a much lower NADPH generation, which is crucial mostly in G6PD-deficient individuals, whose NADPH availability is poor

ABSTRACT

A neurotoxic peptide, granulitoxin (GRX), was isolated from the sea anemone Bunodosoma granulifera. The N-terminal amino acid sequence of GRX is AKTGILDSDGPTVAGNSLSGT and its molecular mass is 4958 Da by electrospray mass spectrometry. This sequence presents a partial degree of homology with other toxins from sea anemones such as Bunodosoma caissarum, Anthopleura fuscoviridis and Anemonia sulcata. However, important differences were found: the first six amino acids of the sequence are different, Arg-14 was replaced by Ala and no cysteine residues were present in the partial sequence, while two cysteine residues were present in the first 21 amino acids of other toxins described above. Purified GRX injected ip (800 µg/kg) into mice produced severe neurotoxic effects such as circular movements, aggressive behavior, dyspnea, tonic-clonic convulsion and death. The 2-h LD50 of GRX was 400 ñ 83 µg/kg

ABSTRACT

The synthesis of analogs of the C-terminal tridecapeptide of gastrin in described. These pseudopeptide analogs were obtained either by replacing the C-terminal phenylalanine amide with 2-phenylethytalcohol or with 2-phenylethylamine, or by replacing the peptid bond between Trp and Leu, or between Leu and Asp with an aminomethylene (CH2NH). The ability of these compounds to stimulate gastric acid secretion in anesthetized rats and to inhibit binding of labeled CCK-8 to isolated cells from rabbit fundic mucosa was tested. [desPhe13, Leu11]-HG-12-I-beta-phenylethylester 33, [desPhe13, Leu11]-HG-12-II-beta-phenylethylester 38 [desPhe13, Leu11]-HG-12-I-beta-phenylethylamide 32, and [desPhe13, Leu11]-HG-12-II-beta-phenylethylamide 37 acted as gastrin receptor antagonists, while [Trp10-((CH2NH)-Leu11]-HG-13-I 31 and (Trp10-((CH2NH)-Leu11]-HG-13-II 36 acted as agonists. Unexpectedly, [Leu11-((CH2NH)-Asp12]-HG-13-I 30 and [Leu11-((CH2NH)-Asp12]-HG-13-II 35 were almost devoid of affinity for the gastrin receptor.

ABSTRACT

The expression of the cellulase transcripts of Trichoderma reesei is controlled by the nature of the energy carbon sources used in the culture medium. Cellulose and the soluble disaccharide sophorose, but not glycerol or glucose, act as inducers. Evidence is presented suggesting that a low constitutive extracellular cellulolytic system catalyzes the formation of a soluble inducer from cellulose, and this inducer triggers the expression of the cellulase transcripts. This basal and cellulose-induced expression of the cellobiohydrolase I mRNAs (cbh1), the major member of the cellulase system, is transcriptionally controlled by two independent cis-acting DNA regions. In addition, expression of the cbh1 transcript is influenced by the physiological state of the mitochondria and this sensitivity is controlled through the 5,-flanking DNA sequence of this gene.

ABSTRACT

Os autores se propoem a relatar o estudo restrospectivo sobre tumor de Wilms, em pacientes admitidos e tratados no Hospital Infantil Albert Sabin, da FUSEC, no periodo de fevereiro de 79 a fevereiro de 83.Neste periodo foram admitidas 16 criancas portadoras dessa patologia, todas submetidas ao tratamento classico preconizado para tumor de Wilms, constando basicamente de cirurgia radical, quimioterapia e radioterapia pos-operatorias. Neste estudo foram aventados e analisados os seguintes parametros: 1 - Correlacao entre a sobrevida e o estadiamento clinico-cirurgico.2Correlacao entre sobrevida e estadiamento histologico.3 - Achados radiologicos (urograficos). 4 - Correlacao entre as condicoes clinicas no momento da internacao e sobrevida. Os autores concluiram que: 1- A sobrevida global nao foi tao satisfatoria como era de se esperar, em funcao dos estadios clinicos avancados e das mas condicoes clinicas iniciais. 2 - O numero relativamente grande de estadios II (de pessimo prognostico) contribuiu para o aumento da mortalidade. 3 - A histologia favoravel demonstrou melhorar o prognostico mesmo em estadios clinicos avancados (sobrevida longa em estadio IV)
